Study of the antitumor effect of inducible nitric oxide synthase in a breast cancer model : analysis of molecular mechanisms

The anti -tumor effect of a lipid A, OM -174 (lipid portion of LPS) was studied in a model of breast cancer in mice. In vivo, OM- 174 increases the survival of mice whereas in vitro it is not toxic to cancer cells. OM -174 binds to TLR4 immune cells inducing the production of cytokines such as IFNγ. In vitro, the combination of IFNγ to lipid A is cytotoxic. The objective of this thesis is to analyze those molecular mechanisms. We have shown both in vitro and in vivo that the cytotoxicity of the lipid A / IFNγ is dependent of TLR4 and of the receptor for IFNγ, and the NOS II expression. Siderophores are strong iron chelators produced by bacteria under iron deficiency conditions. In the present work, we studied the impact of the siderophore pyoverdine, produced by the plant growth promoting rhizobacteria Pseudomonas fluorescens C7R12, on plant physiology from phenotypic to molecular effects with a specific focus on plant growth, immune response and iron homeostasis. Effect of nitrogen nutrition on Medicago truncatula resistance against Aphanomyces euteiches

Nitrogen (N) is a major limiting factor for plant growth. N availability can also impact plant resistance to pathogens by regulating plant immunity. To better understand the links between N nutrition and plant defense, we analyzed the impact of N availability of plant on Medicago truncatula resistance to the root pathogen, Aphanomyces euteiches, taking into account plant genetic variability. This oomycete is considered as the most limiting factor for legume production. Two conditions of N nutrition, non-limiting or deprived in N, and ten plant genotypes were tested in vitro. The production of NO following Arabidopsis treatment witholigogalacturonides (OGs), an endogenous elicitor of plant defense, was assessed using the NOsensitive probe 4, 5-diamino fluorescein diacetate. Des travaux de l’UMR Agroécologie montrent que la réponse immunitaire des plantes est corrélée à leur nutrition en fer et est modulable par la pyoverdine, un sidérophore produit par la souche bactérienne bénéfique Pseudomonas fluorescens C7R12. Le traitement d’Arabidopsis thaliana exposée à une carence en fer par la pyoverdine dépourvue de fer (apo-pyo) restaure en effet la croissance de la plante aux dépens de son immunité. Nous avons émis l’hypothèse que le monoxyde d’azote (NO), une molécule signalétique ubiquitaire, est un acteur de la modulation de la réponse immunitaire des plantes exposées à l’apo-pyo et à sa souche productrice.
